    Ginger Extract Arrests Cell

    Several dietary agents have been shown to arrest the cell cycle, leading to growth inhibition and apoptosis. For example, grape seed proanthocyanidins, green tea polyphenols, epigallocatechin-3-gallate, resveratrol , silymarin/silibinin , genistein , curcumin and ginger affect cell-cycle progression at various stages by specifically modulating cell-cycle-associated proteins. Specific gingerols, such as 6-gingerol and 8-gingerol, have been shown to perturb cell-cycle progression as a chemopreventive strategy. Thus, our next aim was to gain mechanistic insights into GE-mediated antiproliferative activity by determining the specific cell-cycle stage at which GE intervenes. To this end, we examined the cell-cycle distribution profile of GE-treated PC-3 cells by employing a flow cytometric assay using the DNA intercalator dye, propidium iodide. shows the effect of varying GE dose levels on cell-cycle progression of PC-3 cells at 24 h of treatment in a three-dimensional disposition. As shown in , exposure of PC-3 cultures consistently resulted in a statistically significant dose-dependent enrichment of the sub-G1 fraction, suggesting considerable apoptotic cell death.     Because ginger contains several biologically active components, we compared the effects of 6-SHO with those of 6-GIN and 6-PAR in both human and mouse prostate cancer cell lines. As shown in , both 6-GIN and 6-PAR reduced the survival of human and mouse prostate cancer cells. However, each of these compounds was less active on a molar basis than 6-SHO. For example, approximately 4-fold higher concentrations of both 6-GIN and 6-PAR were required to produce equivalent reductions in cell survival as compared with what was seen for 40 mol/L 6-SHO in both the human and mouse prostate cancer cell lines. Western blot analyses with 6-GIN and 6-PAR provided confirmation that pSTAT3Tyr705 was reduced by both compounds at a concentration of 100 mol/L in both LNCaP and DU145 cells. Both 6-GIN and 6-PAR decreased the level of survivin and increased the expression of cyclin-dependent kinase inhibitors p21 and p27 in all three human prostate cancer cells, at the 100 mol/L concentration . TNF-induced phosphorylation of NF-Bp65 and IB was also reduced by 6-GIN and 6-PAR in both DU145 and PC-3 cells at the 100 mol/L concentration . 6-PAR also decreased the phosphorylation of STAT3, NF-Bp65, and the level of survivin in HMVP2 cells . Collectively, these results demonstrate that 6-GIN and 6-PAR have the ability to block growth and reduce survival of both human and mouse prostate cancer cells but that they are both less potent than 6-SHO.
